INTRO
In preparation for testing out some new Manila clam primers (GitHub Issue), Steven asked that I isolate some RNA from some Manila clams we currently have in the tank room (GitHub Issue). However, I remembered we already had some old Manila clam RNA from waaaay back in 2008. The RNA is gill RNA from an experiment where some clams (labeled with S) were spun for 1min in the salad spinner and corresponding controls.
Due to the age, and the fact the RNA had previously been quantified using the NanoDrop (which is not particularly accurate), I opted to quantify the RNA using the Qubit High Sensitivity (HS) RNA Assay.
MATERIALS & METHODS
Samples were quantified using the hsRNA Qubit Assay with the Roberts’ Lab Qubit 3.
Used 1uL of each sample.
RNA was returned to the -80oC in Shellfish RNA Box #3 (Google Sheet)
RESULTS
Qubit data:
- 20261027-rphi-qubit-RNA-hs-gill (Google Sheet)
| Sample_ID | Original sample conc. (ng/uL) |
|---|---|
| RNA Manila clam gill C1 | 89.8 |
| RNA Manila clam gill C2 | 184 |
| RNA Manila clam gill C3 | 158 |
| RNA Manila clam gill C4 | 184 |
| RNA Manila clam gill C5 | 200 |
| RNA Manila clam gill S1 | 70 |
| RNA Manila clam gill S2 | 73.8 |
| RNA Manila clam gill S3 | 88.2 |
| RNA Manila clam gill S4 | 108 |
| RNA Manila clam gill S5 | 130 |
I created a pool of RNA for subsequent use in making cDNA using 1uL of each sample. It was also stored in the same -80oC Shellfish RNA Box #3 (Google Sheet)
